E74-Like Factor 3 Is a Key Regulator of Epithelial Integrity and Immune Response Genes in Biliary Tract Cancer

Abstract The transcription factor E74-like 3 (ELF3) is inactivated in a range of cancers, including biliary tract cancer (BTC). Here, we investigated the tumor-suppressive role ELF3 bile duct cells by identifying several previously unknown direct target genes that appear to be implicated carcinogenesis. directly repressed ZEB2, key regulator epithelial–mesenchymal transition, and upregulated expression CGN, an integral element lumen formation. Loss led decreased cell–cell junctions enhanced cell motility. ALOX5 CXCL16 were also identified as additional targets ELF3; their corresponding proteins 5-lipoxygenase play immune response. Conditioned medium from overexpressing significantly migration natural killer CD8+ T toward conditioned medium. Gene profiling for BTC expressing high levels revealed significant enrichment ELF3-related genes. In xenograft model, activation increased genes, tubular structure tumors, loss vimentin. Overall, our results indicate both epithelial integrity responses BTC. Significance: Thease finding shows regulates host functions tumor suppressor cancer.
